2 General methods and materials: All commercial materials were used as received unless otherwise noted. 1 H and 13 C MR spectra were recorded with Varian Mercury-Plus 400 MR and Varian Mercury- Plus 500 MR spectrometer as solutions ( 1 H 400 or 500 MHz; 13 C 100 or 125 MHz) in CDCl 3 or CD 3 D or d 6 -DMS. Chemical shifts are expressed in parts per million (ppm, δ) and are referenced to CHCl 3 (δ = 7.26 ppm; δ = ppm ) or CD 3 D (δ = 3.31 ppm; δ = 49.0 ppm) or d 6 -DMS (δ = 5.50 ppm; δ = ppm) as an internal standard. All coupling constants are absolute values and are expressed in Hz. The description of the signals include: s = singlet, d = doublet, t = triplet, m = multiplet and dd = doublet of doublets, at = apparent triplet, br = broad. High-resolution mass spectra were measured on an agilent TF-G6230B mass spectrometer. Thin-layer chromate- graphies were done on pre-coated silica gel 60 F254 plates (Merck). Silica gel 60H ( mesh) manufactured by Qingdao Haiyang Chemical Group Co. (China) was used for general chromatography. 2

List of known -arylurea substrates H 2 R + Cl Et 3 DCM, 0 to rt 24 h R H Aniline derivatives (10 mmol) and triethylamine (2.9 ml, 20 mmol) were dissolved in anhydrous CH 2 Cl 2 (10 ml) in a 50 ml three neck round bottom flask. After cooling to 0 C, dimethylcarbamic chloride (1.8 ml, 20 mmol) was added slowly by using a syringe and the mixture was allowed to warm to room temperature and stirred 24 h. After completion, the reaction was diluted with CH 2 Cl 2 (30 ml), washed by sat. ahc 3 (30 ml), 2 HCl (30 ml), brine (20 ml) and dried over MgS 4. The organic solvent was removed by evaporation. Purification by re-crystallization in 3

List of known α-diazo β-keto esters R 1 R 2 + p-absa Et 3 MeC, 0 to rt 24 h R 1 R 2 2 2a-e and 2g-i To a solution of 1,3-dione (10.62 mmol) and p-acetamidobenzenesulfonyl azide (p-absa) (2.5 g, mmol) in anhydrous CH 3 C (30 ml) at 0 C, triethylamine (Et 3 ) (4.43 ml, mmol) was added dropwise. After stirring at room temperature for 24 h, the reaction mixture was concentrated in vacuo. Water (20 ml) was added. The resulting mixture was extracted with diethyl ether (2 20 ml). The combined organic layer was washed with brine (20 ml) and dried over MgS 4. The solvent was removed under reduced pressure, and the residue was purified by a silica gel column chromatography with petroleum ether/ethyl acetate as the eluent to give the entitled diazo compounds 2a-e and 2g-i in 70-90% yields. 5a-c, 7 4

5 P Me + p-absa Me ah / Toluene THF, 0 to rt 24 h 2 2f P Me Me Dimethyl 2-oxopropylphosphonate (3.53 g, 21.3 mmol) was dissolved in 60 ml of dry toluene and ah (0.76 g, 31.9 mmol) was added portion wise, after stirred for 1 h at the 0 0 C, a solution of p-absa (5.0 g, 21 mmol) in 25 ml of dry THF was added dropwise. then, the reaction mixture was stirred at room temperature for 24 h, after the reaction was completed (monitored by TLC analysis), 50 ml petroleum ether was added, then the precipitate was filtered off, and the filter cake was washed with ether (3 x 50 ml), the filtrate was evaporated and the residue was purified by column chromatography on silica gel (PE/EA = 2:1 to 1:2), give the 2f (3.76 g) as yellow liquid, yield 92%. 6 Et 2 + DMF + SCl 2 CHCl 3 H 2 2j Et Thionyl chloride (1.31 ml, mmol) was added dropwise to anhydrous DMF (1.39 ml, mmol) and the mixture heated at 40 C for 2 h. The mixture was concentrated in vacuo to give a solid. The solid was dissolved in chloroform (8.0 ml) and ethyl diazoacetate (3.68 ml, mmol) added at 0 C dropwise during a period of 1 h. The mixture was stirred at room temperature for 16 h. The solvent was removed under reduced pressure and ether (8.0 ml) added. The colorless precipitate was filtered off and the solid dissolved in acetic acid (8.0 ml). The suspension was stirred at room temperature for 16 h. The aqueous solution was extracted with ether (3 30 ml) and the combined extracts were washed with aqueous sodium hydrogen carbonate (20%; 3 30 ml), hydrochloric acid (10%; 3 30 ml), water (3 30 ml) and brine (3 30 ml). The extract was dried over anhydrous MgS 4 and concentrated in vacuo to give the 2j as yellow oil (1.25 g, 49%); 7 The product was used without further purification. 5

6 General procedure for C H activation/cyclization: H R 2 [RhCp * (MeC) 3 ][SbF 6 ] 2 (5 mol%) + R 3 R 4 R DCE, 50 o 1 C,18 h R 1 2 1a-s 2a-j 3a-s and 4a-i To a screw capped vial with magnetic stirrer were added substrate 1a-s (0.15 mmol, 1.0 equiv.), 2a-j (0.225 mmol, 1.5 equiv.), [RhCp*(MeC) 3 ][SbF 6 ] 2 (5 mol%) and DCE (0.5 ml) under air. The reaction mixture was stirred at 50 o C for 18 h. After cooling to room temperature, the resulting mixture was diluted with dichloromethane (DCM) and then filtered through a pad of celite and silica gel. The filtrate was concentrated, and the resulting crude mixture was purified through a silica gel column chromatography to give the corresponding desired products 3a-s and 4a-i. R 2 R 3 R 4 Characterizations of indole products: Methyl 1-(dimethylcarbamoyl)-2-methyl-1H-indole-3-carboxylate (3a) Me 2 Me R f = 0.6, 25% EtAc in Hex; compound 3a was obtained as a colorless oil in 89% yield. 1 H MR (400 MHz, CDCl 3 ) δ (m, 1H), (m, 3H), 3.95 (s, 3H), 3.26 (s, 3H), 2.78 (s, 3H), 2.76 (s, 3H); 13 C MR (100 MHz, CDCl 3 ) δ 166.1, 152.7, 143.7, 134.3, 126.6, 123.5, 122.8, 121.9, 110.6, 106.8, 51.1, 38.4, 36.9, 12.7; HRMS (ESI) calcd for ([M+H] + ), found ([M+H] + ). The combined organic layers were dried over MgS 4, filtered and the solvent was removed under reduced pressure and the residue was purified by column chromatography (silica gel, hexane/ethyl acetate = 3/1) to afford compound 5 as a white solid (0.034 g, 90%). 1 H MR (400 MHz, CDCl 3 ) δ 8.33 (br, 1H), 8.09 (dd, J = 7.6Hz, 1.8H, 1H), 7.31 (dd, J = 7.6Hz, 1.8H, 1H), (m, 1H), 3.94 (s, 3H), 2.75 (s, 3H). This is a known compound reported in the literature. 8 Synthesis of 1H-indole-3-carboxylic acid: A solution of 4i (0.2 mmol), aqueous sodium hydroxide solution (50%, 3 ml) and ethanol (5 ml) was stirred at 80 o C for 24 h. After cooling, 1 HCl (5 ml) was added and the aqueous phase was extracted with EtAc (3 x 10 ml). The combined organic 17
