16S rrna Deep Sequencing for the Characterization of Healthy Human Pharyngeal Microbiome Georgia Gioula, Angeliki Melidou, Panayotis Siasios, Fani Minti, Nikolaos Malisiovas Microbiology Department, School of Medicine, Faculty of Health Science, Aristotle University of Thessaloniki, Thessaloniki, Greece Abstract Background: Deep sequencing methods have made possible the evaluation of the composition of the upper respiratory tract (URT) microbial ecosystem, enabling more accurate studies for the determination of its relationship to human health and disease. The aim of the present study is to characterize the composition and diversity of microbial communities of the pharynx in pharyngeal swabs samples of healthy individuals. Materials and Methods: Ten pharyngeal specimens were obtained from healthy volunteers, who were eligible to the selection criteria. DNA libraries were constructed using primers that selectively amplify the corresponding V2-V9 hypervariable regions of the bacterial 16S rrna. Sequencing was performed using the Ion Torrent PGM instrument. Results: Twelve phyla were identified in the study samples. The most abundant phyla detected, were Firmicutes, Proteobacteria, Bacteroidetes, followed by Actinobacteria and Fusobacteria. Moreover, 79 families, 79 genera and 137 species were identified. In total, 17 unique differentially abundant families, 30 unique differentially abundant genera and 24 unique differentially abundant species were detected among healthy subgroups with adjusted p-values < 0.05. According to the genus level, from the 30 unique differentially abundant genera, Moraxella (Proteobacteria) and Gemella (Firmicutes) were present with a statistical significance in non-smokers (padj= 5.06E-10, and 6.64E-03 respectively). Bifidobacterium (Actinobacteria), Alloscardovia (Actinobacteria), Dialister (Firmicutes) and Filifactor (Firmicutes) were detected with a statistically significant difference mostly in smokers (padj=4.29e-04, 5.76E-04, 1.52E-02 and 5.06E-10 respectively). Conclusions: The URT microbiome varies greatly between individuals. Further studies are needed to better understand its dynamics, the intramicrobiota competition and its interactions with the host in health and disease. Composition of children pharyngeal microbiome by 16S rrna deep sequencing Georgia Gioula, Angeliki Melidou, Panayotis Siasios, Fani Minti, Nikolaos Malisiovas Microbiology Department, School of Medicine, Faculty of Health Science, Aristotle University of Thessaloniki, Thessaloniki, Greece Abstract The upper respiratory tract (URT) is colonised by a large variety of bacteria that constitute the respiratory microbiota. Most do not have a role and may even be protective. The aim of the present study was to analyse the microbiome and characterize the relative abundance of microbial communities of the pharynx by next generation sequencing of the 16S rrna gene of bacteria in healthy children and children with a respiratory infection. Ten phyla were identified in the 8 study subjects. The most abundant phyla detected, were Firmicutes, Proteobacteria and Bacteroidetes, while the relative abundance of each was highly variable across the subjects. At the family, genus and species level, 24 families, 19 genera and 71 species respectively were common both in patients and healthy subjects, while some (28 families and 24 genera) were identified only in healthy subjects and few (7 families, 8 genera and 9 species) were identified only in patients. No statistically significant differences were observed in relation to the age or gender of the subjects. Interestingly, the most abundant bacteria detected in healthy children were Streptococcus, Prevotella, Moraxella, Veillonella and Leptotrichia, while in young patients Moraxella was not detected among the most prevalent bacteria, supporting the notion that it may play a protective role in infection. Protective and pathogenic bacteria have been identified in healthy children and in patients. Such studies can form the basis for new approaches to fight diseases responding poorly to traditional interventions. (Under Submission)

Ειδικά στον αιματοεγκεφαλικό φραγμό έχει προσδιοριστεί ένας τεράστιος αριθμός ειδικών υποδοχέων πρόσδεσης των λιπαρών οξέων.
