Supplementary Methods Immunohistochemistry Five-micrometer-thick sections of formalin-fixed, paraffin-embedded tissue specimens were deparaffinized and dehydrated. All sections were incubated with anti-human ANXA2 rabbit polyclonal antibody (Abcam, Cambridge, UK; dilution 1:1000) and anti-human MSH2 mouse monoclonal antibody (FE11, Dako, Carpinteria, CA; dilution 1:200). Before incubation, the sections were heated for 10 min at 120 o C in an autoclave using sodium citrate buffer at ph 6 and ph 9 (Dako), respectively, for antigen retrieval. Non-specific reactions were blocked with Antibody Diluent (Dako). Both primary antibody incubations were conducted at 4 o C overnight, followed by incubation with EnVision+ Dual link system-hrp (Dako) at room temperature for 30 min. 3.3'-Diaminobenzidine tetrahydrochloride was used as the chromogen. All sections were counterstained with hematoxylin. As a negative control, the primary antibodies were omitted from the reaction sequence. Tissue specimens of T and N samples showing the highest levels of ANXA2 and MSH2 protein expression by the 2DICAL method served as positive controls in all analyses, respectively.
