Supplementary Information As shown in the supplementary figure 1a, the total number of papers that resulted from PubMed search with keyword circulating tumor cells signaling is 1,073 (blue portion of the Venn diagram; 378+648+45+2 publications). This includes the papers that were common to results from keyword searches circulating tumor cells signaling and cancer (648); circulating tumor cells signaling and epithelial mesenchymal transition (2); and results common to all three keyword searches (45). The papers unique to the last two searches were excluded as they were not relevant. After abstract-level screening of these 1,073 papers, 153 were narrowed down for in-depth screening. Similarly, as shown in the supplementary figure 1b, the total number of papers that resulted from PubMed search with keyword circulating tumor cells gene expression is 2,169 (blue portion of the Venn diagram; 488+214+424+1,043 publications). This includes the papers that were common to results from keyword searches circulating tumor cells genes (214); circulating tumor cells expression (1,043); and results common to all three keyword searches (424). The papers unique to the last two searches were excluded due to non-relevance and large numbers. After abstract-level screening of these 2,169 papers, 78 were narrowed down for in-depth screening. Out of these 231 papers (153+78), 98 papers were found to have gene expression data relevant to CTCs from which the gene list was compiled for our analysis. Of these, 93 papers were smallscale qualitative and semi-quantitative experiments that reported expression of a few genes in CTCs. The remaining five papers were based on large-scale transcriptomic experiments (RNA- Seq and Microarray) and reported differential expression of hundreds of genes in CTCs. The final gene list for our pathway analysis comprises mostly of genes from these five papers (7,209 out 7,572 total numbers of genes). These numbers are reported as CTC_ALL (7,572 genes from all 98 papers) out of which 7,209 (CTC_FC) were taken from the five large-scale transcriptomics experiment papers (Supplementary Table S1 and Supplementary Table S2, respectively). The reference information (PubMed IDs; PMIDs) for the 98 papers are listed in Supplementary Table S5. 2
