3 SI3 2. Experimental, chemistry 2.1. General Unless otherwise noted, moisture sensitive reactions were conducted under dry nitrogen. THF was dried with sodium/benzophenone and was freshly distilled before use. Thin layer chromatography (tlc): Silica gel 60 F 254 plates (Merck). Flash chromatography (fc): Silica gel 60, µm (Merck); parentheses include: diameter of the column, eluent, fraction size, R f value. Melting point: Melting point apparatus SMP 3 (Stuart Scientific), uncorrected. MS: MAT GCQ (Thermo-Finnigan); IR: IR spectrophotometer 480Plus FT-ATR-IR (Jasco). 1 H NMR (400 MHz), 13 C NMR (100 MHz): Unity Mercury Plus 400 spectrometer (Varian); in ppm related to tetramethylsilane; coupling constants are given with 0.5 Hz resolution Synthetic procedures (4r)-(3,5-Dihydroxy-4-nitropiperidin-1-yl)-(phenyl)methanone (8b) Nitromethane (0.19 ml, 3.57 mmol) was added to a solution of iminodiacetaldehyde 7b (488 mg, 2.38 mmol) in a mixture of THF (2 ml) and tert-butanol (0.5 ml). NaOH (15 mg, 0.38 mmol) was added and the mixture was stirred for 20 h at rt. The solution was neutralized using Amberlite ion exchange resin (IR-120), filtered and the solvent was removed under reduced pressure. The product was purified by fc ( = 4 cm, h = 16 cm, CH 2 Cl 2 /ethyl acetate = 2/1, V = 30 ml, R f = 0.34 (cyclohexane/ethyl acetate = 2/1)). Colorless solid, mp 135 C (decomposition), yield 316 mg (50 %). C 12 H 14 N 2 O 5 (M = g/mol). 1 H NMR (d 6 -DMSO): δ (ppm) = (m, 1 H, NCH 2 ), (m, 1 H, NCH 2 ), (m, 1 H, NCH 2 ), (m, 1 H, NCH 2 ), (m, 2 H, 2 x CHOH), 4.47 (t, 3 J = 9.8 Hz, 1 H, CHNO 2 ), (m, 2 H, 2 x OH), (m, 5 H, 5 x CH ar ). 13 C NMR (d 6 -DMSO): δ (ppm) = (NCH 2 ), 50.9 (NCH 2 ), 66.9 (CHOH), 67.6 (CHOH), 96.3 (CHNO 2 ), (2 C, C ar ), (2 C, C ar ), (C ar ), (C q ), (C=O). MS (ESI): m/z = 289 [M + Na], 555 [2 M + Na] (4r)-1-Allyl-4-nitropiperidine-3,5-diol (8d) At 0 C allyl bromide (38.5 μl, 0.45 mmol) was added to a solution of nitropiperidinediol 8c (72.1 mg, 0.45 mmol) and K 2 CO 3 (61.5 mg, 0.45 mmol) in THF (2 ml). After 24 h stirring at rt, the solvent was removed under reduced pressure and the product was purified by fc ( = 2 cm, h = 17 cm, cyclohexane/ethyl acetate = 2/1,

4 SI4 V = 10 ml, R f = 0.64 (CH 2 Cl 2 /CH 3 OH = 9/1)). Colorless crystals, yield 14.1 mg (15 %). C 8 H 14 N 2 O 4 (M = g/mol). 1 H NMR (d 6 -DMSO): δ (ppm) = 1.87 (t, 2 J = 10.8 Hz, 3 J = 10.8 Hz, 2 H, 2-CH 2ax, 6-CH 2ax ), 2.92 (dd, 2 J = 11.2 Hz, 3 J = 4.6 Hz, 2 H, 2-CH 2eq, 6-CH 2eq ), 3.01 (d broad, 3 J = 6.4 Hz, 2 H, NCH 2 CH=CH 2 ), (m, 2 H, 2 x CHOH), 4.14 (t, 3 J = 9.8 Hz, 1 H, CHNO 2 ), (m, 2 H, NCH 2 CH=CH 2 ), 5.63 (d broad, 2 H, 2 x OH), (m, 1 H, NCH 2 CH=CH 2 ). Purity (HPLC): 93.2 %, t R = 3.41 min (method CH 3 CN) (4r)-1-Benzyl-4-nitropiperidine-3,5-diol (8e) Trifluoroacetic acid (0.12 ml, 1.52 mmol) was added to a solution of nitropiperidinediol 8c (80.0 mg, 0.30 mmol) in CH 2 Cl 2 (5 ml). The mixture was stirred for 3 h at rt. The solvent was removed under reduced pressure and the residue was dissolved in in CH 3 CN (1 ml). Triethylamine (42 μl, 0.30 mmol) was added to the solution and the mixture was stirred for 10 min. Then benzaldehyde (33 μl, 0.33 mmol) and after 30 min NaBH(OAc) 3 (89.0 mg, 0.42 mmol) were added to the mixture. After 20 h the transformation was stopped by addition of a small amount of water, the solvent was removed under reduced pressure and the product was purified by fc ( = 3 cm, h = 20 cm, CH 2 Cl 2 /ethyl acetate = 4/1, V = 20 ml, R f = 0.20 (CH 2 Cl 2 /CH 3 OH = 10/1)). Colorless resin, yield 39.0 mg (50 %). C 12 H 16 N 2 O 4 (M = g/mol). 1 H NMR (d 6 -DMSO): δ (ppm) = 1.89 (t, 2 J = 10.8 Hz, 3 J = 10.8 Hz, 2 H, 2-CH 2ax, 6-CH 2ax ), 2.89 (dd, 2 J = 11.4 Hz, 3 J = 4.7 Hz, 2 H, 2-CH 2eq, 6-CH 2eq ), (m, 2 H, ArCH 2 ), (m, 2 H, 2 x CHOH), 4.15 (t, 3 J = 9.8 Hz, 1 H, CHNO 2 ), 5.63 (d, 3 J = 6.2 Hz, 2 H, 2 x OH), (m, 5 H, CH ar ). 13 C NMR (d 6 -DMSO): δ (ppm) = 57.6 (2 x NCH 2 ), 60.6 (ArCH 2 ), 68.2 (2 x CHOH), 97.3 (CHNO 2 ), (C ar ), (2 C, C ar ), (2 C, C ar ), (C q ). Purity (HPLC): 97.5 %, t R = 4.57 min (method CH 3 CN).

5 SI (4r)-1-(4-Methoxybenzyl)-4-nitropiperidine-3,5-diol (8f) Trifluoroacetic acid (0.12 ml, 1.52 mmol) was added to a solution of nitropiperidinediol 8c (80.0 mg, 0.31 mmol) in CH 2 Cl 2 (5 ml). The mixture was stirred for 3 h at rt. The solvent was removed under reduced pressure and the residue was dissolved in in THF (1 ml). Then 4-methoxybenzyldehyde (36 μl, 0.30 mmol) and triethylamine (42 μl, 0.3 mmol) were added to the solution. After stirring for 30 min, NaBH(OAc) 3 (89.3 mg, 0.42 mmol) was added and the mixture was stirred for 1 h. The transformation was stopped by addition of a small amount of water and the solvent was removed under reduced pressure. The product was purified by fc ( = 2 cm, h = 15 cm, CH 2 Cl 2 /ethyl acetate = 3/1, V = 10 ml, R f = 0.28). Colorless resin, yield 32.2 mg (38 %). C 13 H 18 N 2 O 5 (M = g/mol). 1 H NMR (d 6 -DMSO): δ (ppm) = 1.85 (t, 2 J = 10.8 Hz, 3 J = 10.8 Hz, 2 H, 2-CH 2, 6-CH 2 ), 2.88 (dd, 2 J = 11.2 Hz, 3 J = 4.6 Hz, 2 H, 2-CH 2, 6-CH 2 ), 3.47 (s, 2 H, ArCH 2 ), 3.73 (s, 3 H, OCH 3 ), (m, 2 H, 2 x CHOH), 4.13 (t, 3 J = 9.8 Hz, 1 H, CHNO 2 ), 5.62 (d, 3 J = 6.2 Hz, 2 H, 2 x OH), 6.89 (d, 3 J = 8.7 Hz, 2 H, 2 x CH ar ), 7.20 (d, 3 J = 8.7 Hz, 2 H, 2 x CH ar ). Purity (HPLC): 94.2 %, t R = 6.31 min (method CH 3 CN) (4aRS,8RS,8aRS)-6-Butyl-8-(pyrrolidin-1-yl)-4a,5,6,7,8,8a-hexahydro-1Hpyrido[3,4-b][1,4]oxazin-2(3H)-one (15d) According to General Procedure A Boc-protected pyridooxazines 14 (115 mg, 0.36 mmol) was reacted with trifluoroacetic acid (0.82 ml, 10.6 mmol) in CH 2 Cl 2 (3.5 ml). Then, alkylation was performed with n-butanal (32.5 μl, 0.36 mmol), NaBH(OAc) 3 (107 mg, 0.5 mmol) and THF (6 ml). The secondary amine was completely converted after 18 h and the product was purified by fc ( = 2 cm, h = 18 cm, CH 2 Cl 2 /CH 3 OH/NH 3 = 9.5/0.47/0.03, V = 10 ml, R f = 0.33). Colorless solid, mp C, yield 75.2 mg (74 %). MS (EM, APCI): m/z = calcd. for C 15 H 28 N 3 O 2 [M + H] , found ; calcd. for C 11 H 21 N 2 O 2 [(M pyrrolidine) + 2 H] , found IR (neat): ν (cm -1 ) = 3197/3066 (w, N-H), 2959/2928/2870 (m, C-H), 1678 (s, C=O) (4aRS,8RS,8aRS)-6-Cyclopentyl-8-(pyrrolidin-1-yl)-4a,5,6,7,8,8a-hexahydro-1H-pyrido[3,4-b][1,4]oxazin-2(3H)-one (15e) According to General Procedure A Boc-protected pyridooxazines 14 (65.0 mg, 0.20 mmol) was reacted with cyclopentanone (18 μl, 0.20 mmol), trifluoroacetic acid (15 μl, 0.20 mmol) and NaBH(OAc) 3 (59.3 mg, 0.28 mmol) in THF (4 ml). After 1 h the amine was completely converted and the product was purified by fc ( = 2 cm, h = 18 cm, CH 2 Cl 2 /CH 3 OH/NH 3 = 9.5/0.47/0.03, V = 10 ml, R f = 0.24 (CH 2 Cl 2 /CH 3 OH = 9/1)). Colorless solid, mp 168 C (decomposition), yield 47.8 mg (81 %). The alkylation was performed with benzaldehyde (65.1 μl, 0.65 mmol),

7 SI7 NaBH(OAc) 3 (127.6 mg, 0.6 mmol) and THF (10 ml). The conversion was complete after 3 h and the product was purified by fc ( = 2 cm, h = 16 cm, CH 2 Cl 2 /CH 3 OH = 9/1, V = 10 ml, R f = 0.30). Colorless solid, mp C, yield mg (90 %). Then alkylation was performed with pyridine-3-carbaldehyde (35 μl, 0.38 mmol), NaBH(OAc) 3 (74.2 mg, 0.35 mmol) and THF (4 ml). The transformation was complete after 20 h and the product was purified by fc ( = 2 cm, h = 16 cm, CH 2 Cl 2 /CH 3 OH/NH 3 = 9.2/0.7/0.1, V = 10 ml, R f = 0.48 (CH 2 Cl 2 /CH 3 OH/NH 3 = 9.5/0.47/0.3)). Pale yellow crystals, mp C, yield 75.5 mg (95 %). Purity (HPLC): 98.5 %, t R = 4.49 min (method CH 3 CN) Butyl (4aRS,8RS,8aRS)-8-(pyrrolidin-1-yl)-2,3,4a,5,6,7,8,8a-octahydro- 1H-pyrido[3,4-b][1,4]oxazine (16d) A solution of AlH 3 (1.33 mmol) was freshly prepared by General Procedure B. Lactam 15d (47.9 mg, 0.17 mmol) dissolved in THF (3 ml) was added dropwise at 0 C. The layers were separated and the aqueous layer was extracted with CH 2 Cl 2 (2 x 3 ml). The combined organic layers were dried (Na 2 SO 4 ) and filtered. The filtrate was concentrated under reduced pressure and the product was purified by fc ( = 2 cm, h = 18 cm, CH 2 Cl 2 /CH 3 OH = 9.8/0.2 CH 2 Cl 2 /CH 3 OH = 9.5/0.5, V = 10 ml), colorless oil, yield 28.5 mg. Since the product contained not converted carbamate 14 (~ 6 %, determined by 1 H NMR-spectroscopy), the mixture was dissolved in CH 2 Cl 2 (2 ml) and trifluoroacetic acid (60 μl, 0.78 mmol) was added. After stirring for 16 h at rt the ph value of the solution was adjusted to ph 7 by addition of a saturated solution of NaHCO 3 and the solvent was removed under reduced pressure. The product was purified by fc ( = 1 cm, h = 16 cm, CH 2 Cl 2 /CH 3 OH = 9.5/0.5, V = 5 ml, R f = 0.58 (CH 2 Cl 2 /CH 3 OH = 9/1)). Colorless crystals, mp 181 C (decomposition), yield 23 mg (68 %). C 19 H 23 Cl 2 N 3 O 3 (M = g/mol). 1 H NMR (CD 3 OD): δ (ppm) = (m, 4 H, N(CH 2 CH 2 ) 2 ), (m, 6 H, N(CH 2 CH 2 ) 2, 7-CH 2, 8-CH), 3.05 (dd, 2 J = 12.9 Hz, 3 J = 10.3 Hz, 1 H, 5-CH 2 ), 3.63 (dd, 3 J = 9.9 Hz, 3 J = 2.6 Hz, 1 H, 8a- CH), (m, 1 H, 4a-CH), 3.77 (d, 2 J = 15.7 Hz, 1 H, ArCH 2 ), 3.96 (d, 2 J = 15.7 Hz, 1 H, ArCH 2 ), 4.06 (d, 2 J = 16.7 Hz, 1 H, 3-CH 2 ), (m, 1 H, 5- CH 2 ), 4.17 (d, 2 J = 16.7 Hz, 1 H, 3-CH 2 ), 4.60 (s, 1 H, NH), (m, 1 H, 7- CH 2 ), 7.22 (dd, 3 J = 8.3 Hz, 4 J = 2.1 Hz, 1 H, 6-CH ar ), 7.45 (d, 4 J = 2.1 Hz, 1 H, 2-
