Conclusions Pembrolizumab is a novel humanised anti-pd-1 antibody In KEYNOTE-001, pembrolizumab showed antitumour activity and had an acceptable adverseevent profile It is the first therapy to deliver superior survival vs. standard of care for PD-L1-positive NSCLC regardless of histology PD-L1 expression in 50% of tumour cells correlated with improved efficacy In KEYNOTE-010 in previously treated patients with PD-L1-positive NSCLC: Pembrolizumab (both 2 and 10 mg/kg Q3W) had superior overall survival to docetaxel This benefit was seen at all PD-L1 levels but was most pronounced in the subgroup of patients with 50% PD-L1 expression Pembrolizumab was associated with less high-grade toxicity than docetaxel These results support pembrolizumab as a potential standard of care for advanced, platinum-pretreated, PD-L1-positive NSCLC
